    藁本药材化学成分、质量控制及药效学研究

    摘要:目的 对藁本药材的化学及药效学进行研究 ,确定藁本药材质量评价的指标并建立评价方法。方法 采用柱色谱的方

    法 ,从中国藁本及辽藁本的根及根茎中共分离得到 17 个化合物。根据藁本药材的功能主治设计了抗炎、镇痛、镇静等药效学

    实验 ,对藁本药材粗提物及单一化合物进行了实验 ,并将实验结果进行了比较 ,确定了能够代表藁本药材功能主治的指标成

    分 ,建立了该指标成分的 HPLC定量分析方法 ,分析了20 个产地的藁本药材。结果 藁本药材中主要成分阿魏酸是能够代表

    藁本药材功能主治的指标成分 ,而且在20 个产地中均含有 ,可以做为评价藁本药材质量的指标成分。结论 中草药的质量控

    制问题是中草药现代化研究的瓶颈问题 ,本实验为此方面的研究提供了可借鉴的方法。

    ABSTRACT:OBJECTIVE To study the chemist ry and pharmacology of Rhizoma and Root L igust icum ,and to find out the con2

    stituents for quality cont rol as well as to establish the analytical method for its quality cont rol .METHOD Seventeen constituents were

    separated and identified f rom medicinal herb Ligusticum by column chromatography. According to the clinical efficacy of genus Ligus2

    ticum ,the pharmacological models of anti2infection ,sedation and analgesia were established. The ext racts of rhizoma Ligusticum and

    constituents were tested on these models and the results were com pared. The indicator for the clinical efficacy of genus Ligusticum was

    found out and the HPLC method for its determination was established. Twenty samples f rom different habitats were analyzed. RE 2

    SULT Ferulic acid was the constituent whose pharmacological activity represented the clinical efficacy of genus Ligusticum and was

    found in twenty samples. It appeared to be suitable as the constituent for quality cont rol of genus Ligusticum. CONCLUSION The

    quality cont rol of Chinese Herb Medicine is the key problem in study of modernization of Chinese Herb Medicine. The paper provides

    the feasible method and idea for the modenization of Chinese Herb Medicine.

    藁本原植物为中国藁本( L igust icum sinense O2

    live)及辽藁本( L . johelenese Nakai et Kitag. ) ,药用

    部位为干燥根及根茎。具有祛风寒、除湿止痛的功

    能。我们两课题组多年来一直从事藁本化学及药效

    学研究 ,对藁本中的化学成分进行了深入研究 ,对乙

    醇提取物及活性成分进行了药效学研究 ,在此基础

    上 ,对主要活性成分进行了定量分析方法的研究 ,并

    制定了 2000 年版中国药典藁本含量测定标准。

    1 藁本化学成分的研究

    从中国藁本及辽藁本的根及根茎中共提取分离

    得到 17 个化合物 ,其中藁本酮[1 ]

    、藁本酚[2 ]

    及新藁

    本内酯[3 ]

    为新化合物。其他化合物均为首次从中

    国藁本及辽藁本的根及根茎中分离得到。从中国藁

    本根及根茎中分离得到的新化合物已报道 ,其他化

    合物的提取分离及结构鉴定见本文 ,从辽藁本的根

    及根茎中分离得到的化合物除佛手柑内酯及孕甾烯

    醇酮外均已报道[3 ]。

    1. 1 仪器与材料

    熔点用 Kofler 熔点测定仪测定 ,温度未校正。

    红外光谱用 Perkin - Elmer 683型红外光谱仪测定。

    核磁共振谱用Bruker - AM500型核磁共振仪测定 ,· 456 · Chi n Pharm J , 2002 September , Vol . 37 No. 9

    色谱用的硅胶 GF254和柱色谱用的硅胶(200～300

    目)以及硅胶 H均为青岛海洋化工厂生产。TLC展

    开剂: ①石油醚2乙醚(80∶ 20) , ②石油醚2丙酮(90∶

    10) , ③石油醚2醋酸乙酯2醋酸(80∶ 20∶ 0. 06) ,显色

    剂为磷酸钅 目和碘。中国藁本及辽藁本经本所宋万

    志教授鉴定。阿魏酸对照品、 β 2谷甾醇对照品、孕甾

    烯醇酮对照品、棕榈酸甘油酯对照品均为自制。

    1. 2 提取与分离

    中国藁本根及根茎 2 kg ,以 95 %乙醇热提 3

    次 ,每次 1 h ,乙醇提取物浓缩至小体积后有白色结

    晶析出 ,过滤后得化合物 Ⅰ结晶(100 g) ,过滤后的

    提取液继续浓缩得乙醇提取物 345 g ......
